摘要: Gonadotropin-releasing hormone-agonist (GnRH-Ag) used in controlled ovarian hyperstimulation (COH) for vitro fertilization and embryo transfer (IVF-ET) has been known to directly affect apoptosis of human cells, but its mechanism is not clearly understood. Therefore, the purpose present study was investigate whether caspase-8, -9, -3 activation poly-(ADP-ribose)-polymerase (PARP) cleavage are involved by which GnRH-Ag induces granulosa-luteal cells. The prospective conducted research institute clinical fertility center university hospital. Human cells collected from IVF-ET patients were cultured treated with 10-6 M or saline as a control. To access terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-digoxigenin nick end-labeling (TUNEL) assay DNA fragmentation analysis preformed 24 h after treatment. Activity examined using fluorogenic substrate. Caspase-8, PARP analyzed Western blot. increased at concentrations over GnRH-Ag. In TUNEL assays, rate apoptotic cell formation treatment significantly compared that (p < 0.05). activity -9 and, investigated substrate only blot analysis, revealed an increase active forms addition, also These results suggest might be induced
