Description of the Retention and Peak Profile for Chromolith Columns in Isocratic and Gradient Elution Using Mobile Phase Composition and Flow Rate as Factors

摘要: The effect of the modifier concentration and flow rate on chromatographic performance a second generation Chromolith® RP-18e column, under isocratic gradient elution with acetonitrile-water mixtures, was examined using four sulphonamides as probe compounds. acetonitrile varied between 5 55% (v/v), 0.1 5.0 mL/min, keeping other factors constant. changes in both retention peak profile were modelled, used to build simple plots, where logarithm factor represented against (in elution, initial concentration), half-widths or widths time at column outlet). A particular plot needed for describing each sulphonamide, but due similar interaction kinetics, unique described all sulphonamides. showed that allegedly Chromolith, deformation observed first applied pressure increasing flow, is decreased.
