Chainia Penicillin V Acylase: Strain Characteristics, Enzyme Immobilization, and Kinetic Studies

摘要: Aerobic cultures of an actinomycete were found to produce penicillin V acylase (PVA) (PA, EC-3.5.1.11) extracellularly. The presence L-2-3 diamino-propionic acid in cell wall and formation sclerotia on culture media led its identification as Chainia, a sclerotial Streptomyces. Partially purified was adsorbed kieselguhr entrapped polyacrylamide gel. immobilized preparation proved effective with respect retention enzyme activity even after 15 successful cycles. pH optimum for crude the range 7.5–8.0, (NH4)2 SO4 fraction it 8.5. showed maximal at 9.5. temperature 55°C. enzyme, ammonium sulfate fraction, Km value 6.13 mM, 14.3 17.1 respectively.
