摘要: Abstract Antibodies are employed ubiquitously in biomedical sciences, including for diagnostics and therapeutics. One of the most important uses is immunohistochemical (IHC) staining, a process that has been improving evolving over decades. IHC useful when properly employed, yet misuse method widespread contributes to “reproducibility crisis” science. We report some common problems encountered with assays, direct readers wealth literature documenting providing solutions this problem. also describe series vignettes include our approach analytical validation antibodies assays have facilitated number biological insights into prostate cancer refutation controversial association viral etiology gliomas. postulate great deal problem lack accuracy stems from awareness by researchers critical necessity end-users validate their laboratories, regardless manufacturer claims or past publications. suggest one reason pervasive end-user research fail realize there two general classes IHC. First, “clinical grade” reagents used pathologists help render diagnoses influence patient treatment. Such diagnostic antibodies, which tend be highly validated prior clinical implementation, vast minority (e.g.  3 800 000), often not extensively commercialization. Given increased problem, both United States, National Institutes Health journals requiring investigators provide evidence specificity antibody-based assays.
