摘要: The bacterium Bacillus subtilis produces the DNA integrity scanning protein (DisA), a checkpoint that delays sporulation in response to damage. DisA scans chromosome and pauses at sites of lesions. Structural analysis showed synthesizes small molecule cyclic diadenosine monophosphate (c-di-AMP). Here, we demonstrate intracellular concentration c-di-AMP rises markedly onset DisA-dependent manner. Furthermore, exposing sporulating cells DNA-damaging agents leads global decrease level this molecule. This drop was associated with stalled complexes halt production increased levels c-di-AMP-degrading enzyme YybT. Reduced cause delay can be reversed by external supplementation Thus, acts as secondary messenger, coupling progression sporulation.
