Diacylglycerol kinase in the central nervous system—molecular heterogeneity and gene expression

摘要: Abstract Diacylglycerol (DAG) is one of the important second messengers, which serves as an activator protein kinase C (PKC). DAG (DGK) phosphorylates to generate phosphatidic acid, thus DGK considered be a regulator PKC activity through attenuation DAG. Recent studies have revealed molecular structures several isozymes from mammalian species, and showed that most are expressed in brain various amounts. We cloned four isozyme cDNAs rat library (DGKα, -β, -γ, -ζ) (previously also designated DGK-I, -II, -III, -IV, respectively) examined their mRNA expressions by situ hybridization histochemistry. Interestingly, it for each distinct pattern brain; DGKα oligodendrocytes, glial cells form myelin; DGKβ neurons caudate-putamen; DGKγ predominantly cerebellar Purkinje cells; DGKζ cerebral cortices. Molecular diversity expression patterns suggest physiological importance enzyme function. Furthermore, functional implications these briefly discussed.
