Wright Department Microbiology Molecular Biology, BYU Master Science Several factors direct the placement specific nucleosomes, which in turn have ability regulate DNA accessibility. These include, but are not limited to, nucleotide sequence preference, modifications, type histones present within nucleosome, presence additional transcription factor or chromatin remodelers. In order contribute understanding complicated processes, three separate hypothesis-driven investigations were carried out. First, we looked into nucleosome positioning phasing closely related cells lines. Second, examined domain level occupancy on various portions chromosome. Finally, generated a novel method that significantly reduces data loss vitro reconstitution experiments caused by fragment-end bias. All our yielded results. examining positions patterns similar cell types find common minor differences types. The may cause unique expression patterns. Secondly, found decreased at chromosome arms is class termed fragile nucleosomes. when recovery applied conservatively dataset, it possible recover 80% lost data.
