摘要: The treatment of mice with high doses 5-fluorouracil (5-FU) results in an enrichment primitive hematopoietic progenitors. Using this procedure, we obtained a new class murine colonies that had very secondary plating efficiencies vitro and could differentiate into not only myeloid cells but also lymphoid lineage cells. phenotypes interleukin-3 (IL-3) induced blast colony were Thy-1-positive lineage-marker-negative. We examined whether these contained stem vivo reconstitute tissues lethally irradiated mice. Blast generate macroscopic visible spleen on days 8 12, 5 x 10(3) sufficient to protect them from irradiation. It was shown 6 or weeks after transplantation cells, donor male detected the thymus female recipients bone marrow by Southern blot analysis using Y-encoded DNA probe. After 10 weeks, partially repopulated In congenic mouse system, donor-derived (Ly5.2) transplantation. Fluorescence-activated cell sorter analyses showed B macrophages developed spleen. thymus, found CD4, CD8 double-positive, single-positive, double-negative populations. Reconstitution delayed These indicate IL-3-induced contain capable reconstituting organs
