摘要: The 3-oxoacyl-CoA thiolases catalyse the last step of fatty acid β-oxidation pathway. In yeasts and plants, this pathway takes place exclusively in peroxisomes, whereas animals it occurs both peroxisomes mitochondria. contrast to baker's yeast Saccharomyces cerevisiae, species from Debaryomycetaceae family also encode a thiolase with predicted mitochondrial localisation. These are able utilise range hydroxyaromatic compounds via 3-oxoadipate which is catalysed by 3-oxoadipyl-CoA presumably work, we studied Oct1p, an orthologue enzyme Candida parapsilosis. We found that cells grown on substrate exhibit increased levels OCT1 mRNA. Deletion alleles impairs growth C. parapsilosis substrates defect can be rescued expression gene plasmid vector. Subcellular localisation experiments LC-MS/MS analysis enriched organellar fraction-proteins confirmed presence Oct1p Phylogenetic profiling revealed intricate evolutionary pattern indicating multiple horizontal transfers among different fungal groups.
