Effect of the regulation of retinoid X receptor-α gene expression on rat hepatic fibrosis

摘要: Aim:  To study the effect of retinoid X receptor-α (RXR-α) expression on rat hepatic fibrosis. Methods:  Rat fibrosis was induced by CCl4, and rats were randomly divided into an early-phase group (2 weeks) a sustained (8 weeks). They then four groups (normal control, fibrosis, negative control RXR-α groups). A recombinant lentiviral vector carrying gene injected to induce intraportal infusion, tissue pathological examination performed, hydroxyproline content detected. Hepatic stellate cells (HSC) cultured in vitro, lentivirus used activate HSC, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) activation assayed detect HSC proliferation. Results: In vivo experiments indicated that group, there significant differences content, RXR-α, α-smooth muscle actin (α-SMA) type I collagen (P   0.05). In vitro studies revealed significantly inhibited α-SMA activated (P < 0.01), as well proliferation (P < 0.01). Conclusion:  The increased degree fibrosis.
