摘要: In this report, we show that the overexpression of tissue transglutaminase (tTG) in human neuroblastoma cell line SK-N-BE(2) renders these neural crest-derived cells highly susceptible to death by apoptosis. Cells transfected with a full-length tTG cDNA, under control constitutive promoter, drastic reduction proliferative capacity paralleled large increase rate. The dying tTG-transfected exhibit both cytoplasmic and nuclear changes characteristic undergoing express high Bcl-2 protein levels as well phenotypic adhesion molecule markers (NCAM neurofilaments) differentiating along neuronal pathway. keeping findings, transfection an expression vector containing segments cDNA antisense orientation resulted pronounced decrease spontaneous retinoic acid (RA)-induced We also present evidence (i) apoptotic program neuroectodermal is strictly regulated RA (ii) apoptosis preferentially occurs substrate-adherent phenotype. For first time, report here direct effect maturation toward These results indicate tTG-dependent irreversible cross-linking intracellular represents important biochemical event induction structural featuring
