摘要: Icariin has been reported to possess high anticancer activity. Colon carcinoma is one of the leading causes cancer-related mortality worldwide. Here, activity icariin against HCT116 colon cells and possible underlying mechanism were studied. The trypan blue staining assay, wound healing clonogenic CCK-8 Annexin V-FITC/PI double method carried out determine changes cell growth migration. mRNA protein expressions determined by quantitative real-time PCR western blot, respectively. Moreover, small interfering RNA (siRNA) plasmid was used examine role p53 in icariin-induced apoptosis cells. significantly suppressed decreasing migration viability, simultaneously promoting apoptosis. exerted anti-tumor effect a dose-dependent manner up-regulating p53. During treatment icariin, p-p53, p21, Bax levels increased, Bcl-2 level decreased. Short time with induced DNA damage Furthermore, cytotoxicity decreased after knockdown or using caspase inhibitors. involved activities caspase-9 caspase-3. repressed line enhancing expression activating functions possibly through Bcl-2/Bax imbalance -3 regulation. also
