Induction of the TNF-alpha promoter in the murine dendritic cell line 18 and the murine mast cell line CPII is differently regulated.

摘要: While it was recently shown that activation of dendritic cells (DC) results in the production a number cytokines, signal pathways and transcription factors involved this process have not been described. To address issue we compared events resulting human TNF-alpha promoter occurring fetal cell line 18 (DC18) with those well-characterized murine mast CPII. As stimuli employed protein kinase C inducer, PMA, Ca2+ ionophore, ionomycin, both which are known to activate large variety intracellular signaling pathways. In DC18 cells, PMA alone induces macrolide-insensitive manner. contrast, CPII, (PMA plus ionomycin) necessary for which, addition, is sensitive immunosuppressive drugs. Mapping showed types so-called kappa factor binding site crucial element induction. We show sequence bound controlled by NF-kappaB proteins p50 (NF-kappaB1) p65 (ReIA), whereas CPII NF-AT AN predominant bind control kappaB promoter. These further experimental data indicate DC, play role and, possibly, other cytokine promoters.
