摘要: Abstract The interferon stimulated gene 15 (ISG15) is strongly induced in many cell types by double-stranded RNA (polyinosinic: polycytidylic acid, poly I:C) and viral infection. In this study, we described the nucleotide, mRNA tissue distribution regulation of an ISG15 from turbot, Scophthalmus maximus (SmISG15). SmISG15 862 bp length, composed two exons one intron, encodes 158 amino acids. deduced protein exhibits highest homology (44.7–71.2% identity) with ISG15s other fishes possesses conserved tandem ubiquitin-like (UBL) domains a C-terminal RLRGG conjugating motif known to be important for functions vertebrates. Phylogenetic analysis grouped into fish ISG15. was constitutively expressed all tissues examined, higher levels observed immune organs. Gene expression performed spleen, head kidney, gills muscle turbots challenged I:C or turbot reddish body iridovirus (TRBIV) over 7-day time course. result showed that upregulated both stimuli four tissues, induction apparently stronger initiated more quickly. A two-wave seen kidney gills, suggesting either IFN-dependent -independent pathway. These results provide insights roles antiviral immunity.
