摘要: Background and Purpose —Growth factors (GFs) may favor the healing of aneurysms treated with endovascular techniques by stimulating neointima formation. Methods —Bilateral carotid were constructed venous pouches in 50 pigs embolized intraoperatively collagen sponges without GFs (platelet-derived growth factor-BB [PDGF-BB] 0.15 or 1.5 μg transforming factor-β1 [TGF-β1] 60 600 ng) each animal. DNA synthesis, cell proliferation, secretion assays performed to assess vitro effects on neointimal cells harvested from aneurysms. 125I-PDGF-BB was used study vivo GF release sponges. The thickness at surface measured 2 weeks after surgery. Since porcine tend heal sponge embolization, this experiment repeated dogs, which have shown a propensity for recurrence same technique, ng TGF-β1 platelet extracts. Results —PDGF-BB stimulated synthesis while strongly increased vitro. Clearance followed biphasic curve, 1.5% exogenous PDGF-BB remaining 1 week. local delivery (0.15 μg) (600 significantly neck aneurysms, had no demonstrable effect. extracts influence canine aneurysms. Conclusions can stimulate formation vivo, but do not compensate deficient thrombosis Effects long-term results embolization remain speculative.
