Effects of 5-azacytidine on the progressive nature of cell transformation.

摘要: C3H 10T1/2 mouse embryo fibroblasts were exposed to 3 microM 5-azacytidine for 24 h and then serially passaged in the absence of examined subsequent changes growth properties. The treated cells showed morphology, saturation density, rate, serum dependence. By 5th passage they acquired ability grow 0.3% agarose, by 30th gave rise fully transformed foci that grew agar, liquid suspension. This progression was rapidly accelerated if cultures derived from 5-azacytidine-treated 48 carcinogen benzo[a]pyrene. Results these studies provide evidence aberrations DNA methylation may be one a series critical events during course multistage carcinogenesis thus enhance evolution tumor cells.
