Rin-like, a novel regulator of endocytosis, acts as guanine nucleotide exchange factor for Rab5a and Rab22

摘要: RIN proteins serve as guanine nucleotide exchange factors for Rab5a. They are characterized by the presence of a homology domain and C-terminal Vps9 domain. Currently three family members have been described analyzed. Here we report identification novel member, Rin-like (Rinl), that represents new interaction partner receptor tyrosine kinase MuSK, which is an essential key regulator neuromuscular synapse development. Rinl localized to synapses but shows highest expression in thymus spleen. preferentially binds nucleotide-free Rab5a catalyzes GDP GTP. Moreover, also GDP-bound Rab22 increases GDP/GTP implicating endocytotic processes regulated Rab22. Interestingly, higher catalytic rate compared closely associated with cytoskeleton thus contributes spatial control signaling at actin-positive compartments. Most importantly, overexpression affects fluid-phase well EGFR endocytosis.
