Diacerein inhibits the pro-atherogenic & pro-inflammatory effects of IL-1 on human keratinocytes & endothelial cells

摘要: We investigated IL-1-induced regulation of genes related to inflammation and atherogenesis in human keratinocytes endothelial cells, if ‘diacerein’, an oral IL-1 inhibiting drug currently approved for use osteoarthritis, would reverse IL-1’s effects on these cells. Primary coronary artery cells were treated with either IL-1α or IL-1β, without diacerein. Using PCR-array, we assessed differential gene-expression regulated by identified 34 pro-atherogenic 68 pro-inflammatory significantly (p<0.05) at least 2-fold IL-1, comparison control. Diacerein completely partially reversed this almost all genes. ELISA, confirmed diacerein’s ability IL-1-driven gene-regulation 11 selected factors, the protein level. The results support a novel idea that diacerein acts as inhibitor IL-1. may have therapeutic applications diminish skin psoriasis attenuate development atherosclerosis. Further investigation into effect inflammation, cardiovascular risk animal models humans is warranted.
