摘要: The purpose of the study was to evaluate efficacy and safety intravitreal pirfenidone for inhibition proliferative vitreoretinopathy (PVR) in a model penetrating ocular injury. Penetrating trauma induced on retina rabbit treated either with 0.1 ml phosphate-buffered saline (PBS) or 0.5% pirfenidone, development PVR evaluated clinically graded after 1 month. Histopathology immunohistochemistry transforming growth factor beta (TGFβ), alpha smooth muscle actin (αSMA), collagen-1 were performed assess fibrotic changes. Expression cytokines vitro-retinal tissues at different time points following PBS injection examined by RT-PCR. Availability vitreous various determined high-performance liquid chromatography pirfenidone. In normal eye, injected any toxic effect. Clinical assessment grading revealed prevention formation pirfenidone-treated animals, gross histology, histopathology confirmed observation. Immunohistochemistry showed expression collagen-I, αSMA, TGFβ eyes compared PBS-treated eyes. Pirfenidone inhibited increased gene observed control could be detected up 48 h eye single injection. did not show adverse effect injection; devoid abnormal clinical sign, intraocular pressure, electroretinography significant change histology remained unchanged. This animal shows that might potential therapy PVR. Further will useful application
