Modulation of HMG-CoA reductase and glutathione-linked enzymes and protection against pro-oxidant induced oxidative damage in colon (Caco-2) cells and rat colon homogenates by phenolic extracts from Shaddock (Citrus maxima) peels

摘要: This study sought to characterize the soluble free and bound phenolic compounds from shaddock (Citrus maxima) peels investigate their effect on 3-hydroxy-methyl-3-glutaryl coenzyme A reductase (HMG-CoA reductase) glutathione-linked enzymes in colon (Caco-2) cells. The radicals scavenging ability protective of extracts against pro-oxidant induced oxidative damage Caco-2 cells rat homogenates were also investigated. phenolics extracted with 80% acetone (v/v), while alkaline (NaOH) acid (HCl) hydrolyzed residue ethyl acetate. HPLC fingerprinting revealed presence gallic acid, catechin, chlorogenic caffeic ellagic epicatechin, rutin, quercitrin quercetin extracts. results that inhibited HMG-CoA activity increased activities glutathione peroxidase peroxyl radical oxidation membrane lipids malondialdehyde production homogenates. Furthermore, scavenged [2,2-diphenyl-1-picrylhydrazyl (DPPH) hydroxyl (OH)] chelated Fe2+ a concentration-dependent manner. showed could serve as dietary means or nutraceutical source for protecting degeneration.
