摘要: Olanzapine (OLZ), an atypical antipsychotic, can be effective in treating patients with restricting type anorexia nervosa who exercise excessively. Clinical improvements include weight gain and reduced pathological hyperactivity. However the neuronal populations mechanisms underlying OLZ actions are not known. We studied effects of on hyperactivity using male mice lacking hypothalamic neuropeptide melanin-concentrating hormone (MCHKO) that lean hyperactive. compared vivo systemic or intra-accumbens nucleus (Acb) administration locomotor activity WT MCHKO littermates. Acute treatment significantly activity, effect is substantially attenuated mice. Furthermore, infusion directly into Acb but To identify contributing mechanisms, we assessed synaptic transmission ex vitro. Intraperitoneal GABAergic neurons. This was also seen vitro by applying to acute brain slices. frequency amplitude more robust than Acb. These findings indicate MCH necessary for hypolocomotor OLZ.
