Localized Changes in Blood-Brain Barrier Permeability following the Administration of Antineoplastic Drugs

摘要: Abstract The effects of four antineoplastic drugs on the permeability blood-brain barrier to Evans blue-albumin and horseradish peroxidase were studied in cats. Extravasation tracer brain tissue was observed only rarely following injection methotrexate, cyclophosphamide, or vincristine. However, 5-fluorouracil (15 mg/kg) caused localized exudation various gray white matter areas 8 13 cats which blue administered 7 hr after drug injection. Electron microscopy revealed that stimulated pinocytotic vesicular transport across capillary endothelial cells possibly it widened tight junctions. Barrier leakage not when time periods longer than elapsed between administration, extravasation occurred once a shorter interval. These results suggest changes bloodbrain administration are reversible fairly short duration. Such may be relevant development secondary intracranial tumors chemotherapy.
