摘要: A structure-validated alignment of 35 extradiol dioxygenase sequences including two-domain and one-domain enzymes was derived. Strictly conserved residues include the metal ion ligands several catalytically essential active site residues, as well a number structurally important that are remote from site. Phylogenetic analyses based on this indicate ancestral enzyme arose single genetic duplication event. Subsequent divergence among dioxygenases has resulted in families, two which substrate preference.
