Alternative splicing of glucokinase mRNA in rat liver.

摘要: The sequences of two near full-length cDNAs encoding rat liver glucokinase are reported. One the is essentially identical to cDNA cloned by Andreone, Printz, Pilkis, Magnuson & Granner. [(1989) J. Biol. Chem. 264, 363-369]. other contains a 151 bp insertion and downstream 52 deletion. inserted block bases has been shown originate from an optional cassette exon, termed 2A, between previously described exons 1 2. conceptual translation product variant mRNA original protein for first 15 amino acids. Next there novel polypeptide sequence 87 residues, comprising 50 residues encoded exon 37 specified altered reading frame in Due deletion, 17 acids reference then missing, after which reverts original. Northern blot analysis with oligonucleotide probes that alternatively spliced represents about 5% total mRNA. Alternative splicing may explain earlier findings minor isoforms hepatic glucokinase.
