摘要: States of phosphorylation neurofilament proteins were examined in the perikarya rat sensory and motor neurons between 3 28 d following either a distal transection [6–7 cm from L4-L5 dorsal root ganglia (DRG)] or proximal (1–2 DRG) sciatic nerve. Paraffin sections right (experimental) left (control) L4 L5 DRG animals with unilateral nerve stained immunocytochemically monoclonal antibodies to phosphorylation- dependent (NF-P), dephosphorylation-dependent (NF-dP), phosphorylation-independent (NF-ind) epitopes on largest (NF200), mid-sized (NF150), smallest (NF68) protein subunits. Increased immunoreactivity NF-P NF200 NF150 was detected experimental DRC at 10 d, peaking by 20 declining near control levels d. Conversely, NF-dP declined beginning 6 reaching maximum decline 10– 16 returning Immunocytochemical changes confirmed biochemical studies tissue homogenates that demonstrated an increase decrease reactivity DRG. Changes immunoreactivities observed only large closely associated chromatolytic these neurons. Marked enhancement chromatolysis, as well NF-dP, occurred (left side) versus (right same animal.(ABSTRACT TRUNCATED AT 250 WORDS)
