Effect of Leukemia Inhibitory Factor on Human Cytotrophoblast Differentiation Along the Invasive Pathway

摘要: PROBLEM : Leukemia inhibitory factor (LIF) is a pleiotropic secreted cytokine that was shown to be essential for blastocyst implantation in mice. Since it well documented LIF produced by the human endometrium, we wondered if this capable of modulating invasive behaviour cytotrophoblastic cells (CTB). METHODS CTB were isolated and purified from first trimester abortions, separated or not into bearing laminin fibronectin receptor (α6β4 α5β1 respectively) using specific monoclonal antibodies magnetic particles. RESULTS We observed rhLIF inhibited secretion gelatinases hCG but remained without effects on fetal (fFN). These exerted different subsets although α6 positive cells, stimulated fFN α5 cells. The effect mainly due its CTB. CONCLUSIONS Taken together these results suggest vitro inhibits differentiation towards an phenotype inhibiting metalloproteinases, increasing deposition extracellular matrix syncytium.
