摘要: Abstract We have generated a monoclonal antibody with specificity for the fifth component of mouse complement (C5). This precipitates two chains C5 from normal serum and inhibits C5-dependent hemolysis in functional test. In this study we describe its application an enzyme-linked immunoadsorbent assay (ELISA assay) detection anti-C5 antibodies serum. Monoclonal coupled to wells ELISA plate specifically binds unfractionated subsequently serves as antigen bind antibodies. By approach circumvented need extensive purification which would be required if was directly plates antigen. Serum C5-immunized mice bound high avidity containing source amounts representing 1 μg 250 ng C5. There no binding C5-deficient source. The immune reaction detected by development enzyme-coupled goat-anti Ig specific various subclasses. method allows qualitative characterization responses is ideal model natural self studies immunological tolerance.
