Endoglin is involved in BMP‐2‐induced osteogenic differentiation of periodontal ligament cells through a pathway independent of Smad‐1/5/8 phosphorylation

摘要: The periodontal ligament (PDL), a connective tissue located between the cementum of teeth and alveolar bone mandibula, plays crucial role in maintenance regeneration tissues. PDL contains fibroblastic cells heterogeneous cell population, from which we have established several lines previously. To analyze characteristics unique for at molecular level, performed cDNA microarray analysis versus MC3T3-E1 osteoblastic cells. followed by validation reverse transcription-polymerase chain reaction immunochemical staining revealed that endoglin, had been shown to associate with transforming growth factor (TGF)-β morphogenetic proteins (BMPs) as signaling modulators, was abundantly expressed but absent knockdown endoglin greatly suppressed BMP-2-induced differentiation subsequent mineralization. Interestingly, did not alter level Smad-1/5/8 phosphorylation induced BMP-2, while it expression Id1, representative BMP-responsive gene. Therefore, is conceivable regulates BMP-2-responsive genes some site downstream phosphorylation. Alternatively, found Smad-2 well phosphorylated BMP-2 cells, knockdown.
