摘要: In this article, we describe our methods and protocols using collision-induced dissociative chemical crosslinking-tandem mass spectrometry (CID-CXL-MS/MS) analysis the practical considerations when implementing these reagents methodology for protein crosslinking studies. The synthesis of novel crosslinkers is described as well their use effectively labeling complexes. Several sample preparation liquid chromatography-tandem are provided including enrichment interpeptide crosslinks. For identification CID-CXL-MS/MS crosslinks, details regarding MS acquisition parameters utilization various spectrometers addressed along with post-data to identify Once approach optimized a target or set targets, it can be used tool biological research studying structure integrated data obtained other techniques, such NMR, X-ray crystallography, cryo-electron microscopy, extended study protein-protein interactions in physiological environments.
