Aging shapes the population-mean and -dispersion of gene expression in human brains

摘要: Human aging is associated with cognitive decline and an increased risk of neurodegenerative disease. We assessed age-related brain gene expression by analyzing the Genotype-Tissue Expression (GTEx) data from 2,522 tissue samples across 13 regions in 169 donors European descent aged between 20 70 years. After controlling for covariates hidden confounding factors, we identified 1,446 protein-coding genes whose one or more correlated anti-correlated chronological age at a false discovery rate 5%. These are involved various biological processes including apoptosis, mRNA splicing, amino acid biosynthesis, neurotransmitter transport. The distribution these among markedly uneven, suggesting variable region-specific response to aging. many genes, e.g., TP37 C1QA, dependent on individuals' genotypic backgrounds. Finally, using dispersion-specific analysis, IL7R, MS4A4E, TERF1/TERF2, differentially dispersed aging, i.e., variances differ groups. Our results demonstrate that region-specific, genotype-dependent, both mean dispersion changes. findings provide foundation sophisticated modeling studies diseases.
