摘要: Some species in the family Ascidiidae accumulate vanadium at concentrations excess of 350 mM, which corresponds to about 107 times higher than that seawater. In these signet ring cells, with a single huge vacuole ion is contained, function as vanadium-accumulating vanadocytes. To investigate mechanism underlying this phenomenon, we performed an expressed sequence tag (EST) analysis complementary DNA library from vanadocytes vanadium-rich ascidian, Ascidia sydneiensis samea. We determined nucleotide sequences 1000 ESTs and BLAST against SwissProt database. found 93 clones metal-related gene homologues, including ferritin heavy subunit, hemocyanin, metallothionein. Two ESTs, particular, exhibited significant similarity vanabins have been extracted A. samea blood cells low molecular weight vanadium-binding proteins. named genes encoding vanabin3 vanabin4. Immobilized metal affinity chromatography revealed novel vanabin homologues bind vanadium(IV) ions.
