摘要: Genetic inheritance plays a significant role in inter-individual variation of drug response (efficacy and toxicity). Pharmacogenetics pharmacogenomics are the studies using genetic variations to predict response. The study methodology can be grouped into three approaches: candidate gene approach, pathway-based analysis, genome-wide analysis. approach has produced some striking examples pharmacogenetic markers, but it also resulted numerous inconsistent associations. which assesses combined effects panel polymorphisms that interact same pathway and/or different pathways, may amplify individual enhance predictive power. Both hypothesis-driven, relying on priori knowledge polymorphism functions. In contrast, scan is hypothesis-generating discovery-driven no prior disease or action mechanisms required. this chapter, we highlight major accomplishments challenges these approaches discuss pros cons each.
