Plasma collected from heparinized blood is not suitable for HCV-RNA detection by conventional RT-PCR assay

摘要: Detection of hepatitis C virus-RNA (HCV-RNA) in serum or plasma is considered a reliable marker for ongoing HCV infection. HCV-RNA was measured routinely collected from heparinized blood patients with chronic and varying results were found. In this study, detected peripheral mononuclear cells (PBMCs) 16 out 17 anti-HCV positive C. contrast, found only one these seventeen patients. The addition heparin to known containing solutions before reverse-transcription separately PCR, demonstrated the inhibitory effect on reverse transcription amplification reaction. Furthermore, evidence provided that PCR should be preferably into EDTA vacutainers.
