摘要: Infection of cells by human papillomaviruses (HPVs) associated with malignant genital lesions has not been studied because the lack an in vitro system and unavailability virions. We have now used virus-like particles (VLPs) HPV type 33 to analyze initial events interaction capsid cell lines. Binding VLPs HeLa was observed biochemical assays immunofluorescence. VLP binding inhibited antisera raised against but monoclonal antibodies recognizing either L1 or L2 epitopes accessible on VLPs. Under saturating conditions, approximately 2 x 10(4) were bound per cell, a dissociation constant about 100 pM. composed alone as well both proteins, indicating that is required for binding. dissociated into capsomers did bind, demonstrating intercapsomer contacts are required. Neither nor simian virus 40 virions competed Uptake small smooth endocytic vesicles demonstrated immunoelectron microscopy. Cellular sensitive trypsin sialidase, N-glycosidase, octyl-beta-D-glycopyranoside treatment, suggesting surface protein involved Cell lines originating from variety tissues organisms distantly related insects humans similar efficiency specificity. Therefore, putative receptor mediating attachment should be highly conserved cannot responsible species tissue specificity HPVs.
