Nitriles form mixed-coligand complexes with 99mTc-HYNIC-peptide

摘要: Abstract Using a 12-amino acid peptide conjugated with HYNIC as model, we investigated nitriles possible coligands for labeling 99m Tc. After the preparation of Tc labeled HYNIC-peptide using tricine coligand, addition acetonitile was found by reverse phase HPLC to block further coligand exchange ethylenediamine diacetic (EDDA) at room temperature. The this nitrile changed pharmacokinetics in normal mice towards faster clearance and significant differences accumulation most tissues sampled. By replacing acetonitrile cyanoacetate, not present eluant, it show existence new, more hydrophilic, species HPLC. We conclude that can act HYNIC-conjugated peptides tricine. Furthermore, presence during Sep-Pak or purification may inadvertently generate mixed tricine/acetonitile Tc-HYNIC-peptide complex.
