Etazolate (SQ20009): electrophysiology and effects on [3H]flunitrazepam binding in cultured cortical neurons

摘要: The actions of etazolate (SQ20009) on cultured cortical neurons have been examined in electrophysiological experiments and receptor binding studies. Etazolate (0.3 to 100 microM) prolongs the duration spontaneously occurring IPSPs. Higher concentrations produce an increase membrane chloride conductance, effect which is picrotoxinin bicuculline sensitive. potentiates response exogenously applied GABA acts synergistically with diazepam enhance GABA-mediated conductance. does not glycine-mediated conductance changes. increases [3H]flunitrazepam stimulates enhancement binding. In addition, etazolate-mediated are Cl- dependent dose relationships for effects similar physiological These data suggest that interacts postsynaptic complex at a site distinct from either recognition or benzodiazepine inhibition
