Quantification of [1-(5-fluoropentyl)-1H-indol-3-yl](naphthalene-1-yl)methanone (AM-2201) and 13 metabolites in human and rat plasma by liquid chromatography-tandem mass spectrometry.

摘要: Abstract AM-2201 is a popular synthetic cannabinoid first synthesized in 2000. pharmacokinetic and pharmacodynamic data are scarce, requiring further investigation. We developed sensitive method for quantifying 13 metabolites plasma to provide tool metabolic, studies. Analysis was performed by liquid chromatography-tandem mass spectrometry. Chromatographic separation gradient elution on biphenyl column with 0.1% formic acid water/0.1% acetonitrile:methanol 50:50 ( v/v ) mobile phase. Sample preparation (75 μL) consisted of an enzymatic hydrolysis supported extraction. The validated human 0.025 or 0.050–50 μg/L working range, cross-validated rat plasma. Analytical recovery 88.8–110.1% target concentration, intra- (n = 30) inter-day imprecision  N -(5-hydroxypentyl), JWH-018 -pentanoic concentrations were 4.8 ± 1.0, 0.15 ± 0.03, 0.34 ± 0.07 μg/L, respectively, 8 h after administration at 0.3 mg/kg (n = 5).
