Mitotic recombination in germ cells generated two major histocompatibility complex mutant genes shown to be identical by RNA sequence analysis: Kbm9 and Kbm6.

摘要: Abstract RNA sequencing represents a major procedural simplification for nucleotide sequence analysis of transcribed gene. Using newly adapted mRNA and cDNA procedures, we have sequenced 855 nucleotides Kbm9 mRNA, corresponding to the codons aminoterminal 285 amino acids. The inferred DNA gene differs from parental Kb by single alterations in each 116 121, resulting Tyr----Phe Cys----Arg substitutions, respectively. is identical that another independently arising MHC mutant gene, Kbm6. As both Kbm6 genes were generated recombination between Q4 genes, our data indicate genetic interactions occurred at least twice. relatively large extent identity may be responsible frequent two genes. parents original bm9 mice had five offspring, which can explained mitotic germ cells, producing gonadal mosaicism C57BL/6 mother. Thus, recombination, not meiotic appears formation some mutants. Such mechanism probably plays role generation diversity histocompatibility complex.
