Anti-inflammatory sesquiterpene lactones from Tithonia diversifolia trigger different effects on human neutrophils

摘要: The tagitinins isolated of Tithonia diversifolia (Hemsl.) A. Gray, Asteraceae, are the most studied sesquiterpene lactones due to their wide spectrum pharmacologic activities, especially related with nuclear factor-kappa B inhibition. Nevertheless, detailed studies about mechanism action its active compounds still lacking. Neutrophils perform a fundamental role in inflammatory response several etiologic factors. However, effect on human neutrophil is not yet clearly known. We investigated tagitinin C (1), F (2) and A (3) activation survival neutrophils establish possible effects mechanisms inflammation. Human were purified from peripheral blood cultivated presence or Escherichia coli lipopolysaccharide. enzymatic activity, apoptosis secretion cytokines rate determined after 18 h. Lipopolysaccharide-induced myeloperoxidase activity was significantly inhibited only by (2). Apoptosis increased it occurred independently lipopolysaccharide dexamethasone. Tagitinins decrease lipopolysaccharide-induced interleukin-6, interleukin-8 Tumor necrosis factor alpha production neutrophils. Together, these results indicate that exhibit anti-inflammatory lactone decreased products without inducing apoptosis.
