Extending particle tracking capability with Delaunay triangulation

摘要: Particle tracking, the analysis of individual moving elements in time series microscopic images, enables burgeoning new applications, but there is need to better resolve conformation and dynamics. Here we describe advantages Delaunay triangulation extend capabilities particle tracking three areas: (1) discriminating irregularly shaped objects, which allows one track items other than point features; (2) combining space connect missing frames trajectories; (3) identifying shape backbone. To demonstrate method, specific examples are given, involving analyzing time-dependent molecular conformations actin filaments λ-DNA. The main limitation this shared by all clustering techniques, difficulty separate objects when they very close. This can be mitigated inspecting locally remove edges that longer their neighbors also link two using methods described here, so combination with edge removal robustly applied processing large data sets. As common software packages, both commercial open source, construct on command, paper computationally efficient easy implement.
