Functional Recovery in Hemiparkinsonian Primates Transplanted with Polymer-Encapsulated PC12 Cells

摘要: Cross-species neural grafting of cell lines immunoisolated by a permselective polymer membrane represents promising source neuroactive molecules for the treatment neurodegenerative diseases. Utilization line xenogeneic origin is advantageous since transplanted cells will be rejected host immune system in case breakage immunoisolating envelope. Polymer-encapsulated PC12 cells, dopaminergic derived from rat pheochromocytoma, were five Macaca fascicularis monkeys which had been previously rendered hemiparkinsonian unilateral carotid injection 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine. Well-preserved, tyrosine hydroxylase positive encapsulated observed lesioned striatum up to 5 months after implantation. Four out received polymer-encapsulated showed significant behavioral improvement, whereas three implanted with either bovine chromaffin or empty envelopes no amelioration.
