摘要: The mononuclear lymphoid cell population in human pregnant uterus mucosa, decidua, from early normal pregnancies was studied phenotypically and functionally. phenotype determined situ by immunohistochemistry, isolated decidual preparations immunofluorescence flow cytometry. A mild isolation procedure of gentle mechanical disruption followed density gradient centrifugation used. Leukocytes comprised a large part the tissue. They were present aggregates mainly situated adjacent to glandular epithelium. In addition, individual leukocytes intraepithelially, as well scattered between stromal cells around vessels lacunes. Four lymphocyte populations approximately same size identified: TCR gamma delta+/CD56+ cells, delta+/CD56- delta-/CD56+ alpha beta+/CD8+ cells. delta- expressing about 60% T CD4-/CD8-, half delta+ expressed memory/activation marker CD45RO. Kp 43 Ag, earlier described on activated CD56+ peripheral blood, essentially only decidua. At least 50% beta+ CD8+. function(s) either one these might be prevent immunologic reactions against fetus, protect unwanted extensive invasion trophoblasts, or uteroplacental unit infection. Decidual did not respond stimulation alloantigens mitogenic anti-CD3 mAb but responded extent PBMC lectins. surface TCR/CD3 complex low freshly lymphocytes, could up-regulated upon with PMA/Ionomycin. Local selective down-regulation expression activation involving this mechanisms which maternal reaction semiallogeneic fetus is prevented.
