Isolation and cultivation of mesenchymal stem cells from iliac crest bone marrow for further cartilage defect management.

摘要: Aim to validate isolation and cultivation methods of bone marrow mesenchymal stem cells (BM-MSCs) from iliac crest, compare biological characteristics BM-MSCs different age groups for preparation autologous cell therapy in cartilage defect. Methods patients undergoing spinal surgery were selected grouped according age. Iliac crest the was aspirated. isolated then cultivated. Their including morphological appearances surface biomarkers evaluated. Growth curves observed. Sterility Mycoplasma tests also performed quality assessment culture procedure. Results average, cultivated-BM-MSCs reached number 7.56-22.95 x 106 4-7 weeks period. all showed same morphology, shape (CD105+, CD73+, CD34-, CD45-, CD14-, CD19-, HLA-DR-). Conclusion our procedures isolating cultivating have required amount implantation into lesion. In addition, cultivated-BM-MSCs' accordance with International Society Cell Therapy (ISCT) MSCs criteria.
