摘要: Abstract Chansu, a galenical preparation of the dried white venom Chinese Bufo bufo gargarizans, is one major components Kyushin, traditional medicine. Kyushin reported to have cardiotonic effect that has been suggested be due action bufadienolides such as bufalin and cinobufagin. Recently, we found administration in male rats diminished luteinizing hormone (LH) response gonadotropin-releasing (GnRH) secretion testosterone both vivo vitro. These observations suggest Chansu may possess hypogonadal effects rats. In present study, methanol extract on hypothalamic-pituitary-testicular function were examined. Crude was extracted by purified Sep-Pak C18 column. No activity bufalin, cinobufagin, estradiol, or digoxin detected; all used this study extract. A single intravenous injection resulted decrease basal (20% 55%) human chorionic gonadotropin (hCG)-induced (35% 40%) levels plasma GnRH-induced level LH (25% 30%). Administration vitro decreased hCG-stimulated production 60% 70% 40% 60%, respectively, well spontaneous forskolin- 3-isobutyl-1-methylxanthine (IBMX)-induced accumulation adenosine 3′,5′-cyclic monophosphate (cAMP) 30% 45% rat testicular interstitial cells. Although release anterior pituitary glands diminished, GnRH mediobasal hypothalamus enhanced administratio results bufalin-free extracts inhibit rats, part, (1) cAMP, (2) gonadotropin, (3) reduction responses GnRH.
