摘要: The disease caused by Tomato leaf curl New Delhi virus (ToLCNDV), which is naturally transmitted the whitefly Bemisia tabaci, causes important economic losses in cucurbit crops. availability of simple and efficient inoculation protocols detection methods necessary for screening varieties germplasm collections as well breeding populations. We evaluated infectivity ToLCNDV inocula prepared using three different buffers mechanical sap a susceptible variety zucchini. found that inoculum with buffer III, contains polyvinylpyrrolidone, highly inoculation, 100% plants displaying severe symptoms 21 days post-inoculation. Using this buffer, we mechanically inoculated 19 commercial crops (six cucumber, six melon seven zucchini), evolution diagnosed infection nine (four based on serology, four molecular hybridization one PCR detection). results revealed all are susceptible, although cucumber display less than those or All were (more 85% testing positive) zucchini, but percentage positive detected serology ranged from 20.4% Squash (SLCV) antiserum, to 78.5% DNA extract hybridization. Overall, best obtained PCR, 92.6%, 92.4% 98.4% zucchini plants, respectively, positive. When considering overall crops, Watermelon chlorotic stunt (WmCSV) antiserum extract, respectively. methodology developed information great relevance selection tolerant resistant ToLCNDV.
