摘要: A broadband hyperspectral imaging (HSI) technique, originally developed for remote sensing and in geophysical applications, recently has been employed to enable noninvasive label-free optical of biological specimens. Here we report our results on microscopy non-labeled erythrocytes study the distribution hemoglobin. We have chosen focus cellular hemoglobin because it is an endogenous chromorphore that present high concentrations, which makes a good candidate establishing proof principle. In data cubes (x, y, λ) are collected by either wide-field or confocal microscope equipped with light source, such as white laser spectral engine narrow-band selection wavelength scanning achieved fast optoelectronic devices. The obtained then analyzed algorithms based angle mapper, yielding set unique absorption signatures (i.e. endmembers) corresponding subforms hemoglobin, oxyhemoglobin methemoglobin. Wavelength-dependent scattering cell membranes also resolved. Unique endmembers specific identified used build map intracellular estimating abundance each from cube.
