摘要: We introduce a precise three-dimensional (3D) localization method of spherical gold nanoparticle probes using model-based correlation coefficient mapping. To accomplish this, stack sample images at different z-positions are acquired, and 3D intensity profile the probe serving as model is used to map out positions nanoparticles in sample. By this imaging method, can be achieved techniques with complicated point spread functions (PSF) such differential interference contrast (DIC) microscopy. demonstrated precision 4–7 nm laterally 16 axially for 40-nm nanospheres an rate 10 frames per second.
