摘要: Publisher Summary This chapter examines the recent progress with isolation of biochemically defined mutants altered in phospholipid biogenesis, especially use enzyme-specific autoradiography colony preparations immobilized on filter paper. Most existing have been isolated from bacterium E. coli , although extensions techniques to mammalian cells grown tissue culture are also considered. A classical approach obtaining a metabolic pathway involves auxotrophs requiring certain end products or intermediates for growth. In case metabolism, it is possible feed early precursors, such as fatty acids glycerol-3-phosphate. The success brute force means isolating undisputable, but actual assay and mapping by extremely tedious, if more than one two enzymes be examined. For this reason, rapid screening assays detecting lipid directly developed.
