摘要: IL-15 is a pleiotropic cytokine with IL-2-like functions. As was shown to be mitogenic for T cells, we wondered whether human blood-derived dendritic cells (DC), as the primary stimulators of cell responses, are able produce IL-15. To test our hypothesis, DC were grown under serum-free conditions from peripheral blood using granulocyte-macrophage CSF and IL-4. Cultures assayed mRNA production at various times by semiquantitative reverse transcription-PCR. Low baseline signals detected days 0 5 culture. A significant increase 9 When further enriched immunomagnetic beads >98% purity determined CD83 staining, exclusively found in CD83+ fraction. This paralleled protein release 12 CTLL-2 assay ELISA. In addition, levels increased >10-fold adding paramagnetic cultures, thereby inducing phagocytic activity. Furthermore, supernatants tested chemokinetic chemotactic activities checkerboard filter assay. It that express activity cells. blocked almost completely addition an anti-IL-15 mAb. Our data show contain functional induced Protein triggered DC-derived has suggesting role attractant during initial DC/T interaction.
