PGD for monogenic disorders: aspects of molecular biology

摘要: Preimplantation genetic diagnosis (PGD) for monogenic diseases has known a considerable evolution since its first application in the early 1990s. Especially technical aspects of itself, single-cell analysis, constantly evolved to reach levels accuracy and efficiency nearing those on regular DNA samples. In this review, we will focus molecular biological techniques that are currently use most advanced centers PGD disorders, including multiplex polymerase chain reaction (PCR) post-PCR diagnostic methods, whole genome amplification (WGA) multiple displacement (MDA). As it becomes more clear when comes ethically difficult indications, goes further than prenatal (PND), also briefly discuss ethical issues.
